Hi-C is a powerful tool for understanding 3D chromosome architecture, how chromatin topology influences cellular function and how topological changes contribute to disease. However, traditional Hi-C, relying on sequence biased restriction enzymes, has significant limitations. The Dovetail® Micro-C Assay addresses many of these limitations.
The MNase Advantage
The Dovetail Micro-C Kit utilizes micrococcal nuclease (MNase), generating highly uniform fragments and uniquely capturing nucleosome positioning and an even coverage across the genome. The result? Increased mapping resolution aligned with nucleosomes. Micro-C detects higher-order features, such as chromatin loops, using proximity ligation data by enriching long-range informative reads and capturing chromatin interaction frequency.
With only requires 800 M reads, Dovetail® Micro-C Kit generates 1 kb matrices. Compared to the 1.2-1.6 billion reads required by restriction enzyme-based Hi-C methods, we greatly decrease sequencing costs.

Explore the data
The contact matrices to the right show an enrichment of contacts compared to traditional Hi-C approaches due to Micro-C’s greater signal-to-noise. All matrices were generated from 800 million total read pairs and contacts were balanced.
Micro-C demonstrates increased sensitivity for topological features at lower sequencing depths. The difference is visible to the naked eye.
Micro-C produces more uniform coverage of the genome
The genome coverage map to the left compares different enzyme approaches and sequencing coverage. The Micro-C assay overcomes sequence gaps present in traditional Hi-C data because Micro-C is not limited by low restriction site density.
The MNase-based approach of the Micro-C assay captures accurate and non-biased chromatin topology across the entirety of the genome.
